Abstract: To identify the pathogenic bacteria causing the death of a Vulpes zerda from respiratory failure at a zoo in Guangdong Province,lung tissue samples were collected for bacterial isolation and purification,morphological microscopy,biochemical tests,antimicrobial susceptibility testing,16S rRNA gene sequencing,phylogenetic tree construction and virulence gene detection. Results showed that one strain of Trueperella pyogenes(a Gram⁃positive, short⁃rod⁃shaped bacterium)was isolated from the lung tissue. Biochemical tests indicated that the isolate was positive for lactose,maltose,and glucose fermentation,but negative for sucrose,rhamnose,and mannitol utilization. 16S rRNA gene sequencing revealed more than 99% homology between the Vulpes zerda isolate and reference strains of T. pyogenes from swine and cattle,clustering within the same branch on the phylogenetic tree. Virulence gene detection confirmed the presence of at least six virulence genes:Plo ⁃ g2,Plo ⁃ g1,NanP,FimA,FimE,and FimC. Antimicrobial susceptibility testing demonstrated sensitivity to multiple antibiotics tested,with resistance observed only to two sulfonamide antibiotics(sulfamethoxazole and trimethoprim). This study enriches the characterization of Trueperella pyogenes derived from Vulpes zerda,providing a reference for research on wildlife pathogens

摘要: 为确定广东省某动物园耳廓狐(Vulpes zerda)因呼吸衰竭死亡的致病原,采集其肺脏 组织进行细菌分离纯化、形态镜检、生化试验、药敏试验、16S rRNA基因序列比对及系统进化树 构建以及主要毒力基因检测。结果,在肺脏组织中鉴定出1株革兰阳性、短棒状的化脓隐秘杆 菌(Trueperella pyogenes);生化试验显示分离菌对乳糖、麦芽糖和葡萄糖等呈阳性,对蔗糖、鼠李 糖和甘露醇等呈阴性。16S rRNA基因测序比对显示耳廓狐源分离菌与已报道的猪源、牛源化 脓隐秘杆菌参考菌株同源性>99%,在进化树上处于同一分支。毒力基因检测发现该分离株9 种毒力基因共检出6种(Plo?g2、Plo?g1、NanP、FimA、FimE、FimC)。药敏试验显示该菌对所测试 的多种抗菌药物具有敏感性,仅对两种磺胺类药物(磺胺甲恶唑、甲氧苄啶)表现为耐药;该试 验丰富了耳廓狐源化脓隐秘杆菌的相关特征,为野生动物病原学研究提供了参考。
