体细胞核移植技术在蓝塘猪遗传资源保护中的应用

摘要: 该试验旨在通过体细胞核移植技术对蓝塘猪进行克隆,为我国优良地方猪种或濒危猪种的遗传资源保护提供新的有效途径。试验通过采集蓝塘猪耳组织进行体细胞克隆,比较了组织块法和胶原酶消化法对蓝塘猪体细胞建系的影响,并对比分析了不同性别的蓝塘猪供体细胞制备的克隆胚胎在体外培养的发育效果,最后进行了克隆胚胎的体内移植。结果表明,与组织块法相比胶原酶法能更快的获得细胞及更多的细胞总量。不同性别的克隆胚胎在体外培养的卵裂率和囊胚细胞总数之间无显著差异,但雄性胚胎的囊胚率显著高于雌性(P<0.05)。通过3个批次的蓝塘猪克隆胚胎体内移植,最终成功获得4头健康克隆仔猪。该研究利用体细胞核移植技术成功获得了国内首例体细胞克隆蓝塘猪,为蓝塘猪遗传资源保护提供了可靠的技术手段。

Abstract: This study aimed to clone the Lantang pig using somatic cell nuclear transfer technology,offering a novel and efficient approach for preserving the genetic resources of exceptional local and endangered pig breeds in China. We compared the tissue block method and collagenase digestion method to establish Lantang pig somatic cell lines. Additionally,we analyzed the developmental outcomes of cloned embryos created from Lantang pig donor cells of different genders in vitro and transplanted them in vivo. The results indicated that the collagenase method yielded cells more rapidly and in higher quantities compared to the tissue block method. While there were no notable differences in cleavage rates and total blastocyst cell counts between cloned embryos of different genders in vitro,the blastocyst rate of male embryos was significantly higher than that of female embryos(P<0.05). Through in vivo transfer of cloned embryos from Lantang pigs in three separate batches,we successfully obtained four healthy cloned piglets. This study marks the first successful case of somatic cell cloning of the Lantang pig in China,providing a dependable technical tool for preserving the genetic resources of this breed.
